Abstract

Water apple (Syzygium aqueum) is known to contain flavonoid compounds that exhibit antioxidant activity. The leaves of water apple are part of the plant that possess medicinal properties due to the presence of flavonoids, phenols, terpenoids, and tannins, which are beneficial for maintaining and improving physiological systems as well as preventing diseases. This research aimed to investigate influence of different solvent types on total flavonoid content in water apple (Syzygium aqueum) leaves. The extraction of water apple leaves was performed using maceration method and different solvents based on their polarity, such as methanol (polar), ethyl acetate (semi-polar), and n-hexane (non-polar) with a ratio of (50 grams: 500 ml). The resulting extracts underwent several tests, including microscopic examination, macroscopic examination, color test, thin-layer chromatography, and UV-Vis spectrophotometry. The results of this study indicated that there was an effect of different types of solvents on the total flavonoid content of water apple leaves. Among three types of solvents tested, methanol yielded the best results, with a thin-layer chromatography (TLC) Rf value of 0.38 and an HRf of 38. Additionally, based on UV-Vis spectrophotometry analysis, the total flavonoid content was found to be 5.66%. The conclusion of this study is that methanol solvent is the highest compared to other solvents in producing total flavonoid content of water guava leaf extract (Syzygium aqueum).
